510 Complex phototoxic properties of a cigarette smoke extract on human keratinocytes

چکیده
Sun exposure and cigarette smoke, studied separately, are two well-known environmental factors that accelerate skin aging. However, their combined effects has been poorly characterized. Some studies have determined smoke components can accumulate in the by contact or systemic effect after inhalation. Moreover, it is well-established solar rays penetrate epidermis dermis. Those could thus interact with one another skin. The aim of this study to assess phototoxic on skin, more precisely A extract (CSE) was obtained capturing soluble fraction smoke. performed a simulator at 14.5 29 kJ/m2 UVA, representing 15 30 minutes sun its zenith. CSE phototoxicity human keratinocytes using an MTS assay, establishing cellular metabolic activity. photo-oxidation fluorescent markers measuring ROS levels. total antioxidant capacity (TAC) also assessed. irradiation alone showed no cytotoxicity up maximal doses tested. highly when irradiated. Indeed, exposed 5% respectively 39±1% 2±17% viability (p-value<0.001). Photo-oxidation results indicate decrease type I slight increase II ROS. TAC test revealed capacities. Our synergistic toxicity between cells, caused part reaction. work still focussing deciphering mechanisms involved synergy.

Effect of cigarette smoke extract on arteriolar dilatation in vivo.
The goal of this study was to determine whether cigarette smoke extract alters dilatation of arterioles in vivo in response to agonists that produce activation of ATP-sensitive potassium channels and activation of adenylate cyclase. By using intravital microscopy, we measured diameter of arterioles contained within the microcirculation of the hamster cheek pouch during suffusion with agonists i...

Maternal smoking has been recognized as a common cause of low birth weight, preterm birth and the decrease of gestational age period. Unfortunately, there is an increasing interest within public especially woman in Iran in the tobacco products consumption. On the other hand, the deleterious effect of maternal smoking on human fetus in pregnancy period especially in the first trimester encourage...
متن کاملEffects of cigarette smoke extract on human airway smooth muscle cells in COPD.
We hypothesised that the response to cigarette smoke in airway smooth muscle (ASM) cells from smokers with chronic obstructive pulmonary disease (COPD) would be intrinsically different from smokers without COPD, producing greater pro-inflammatory mediators and factors relating to airway remodelling. ASM cells were obtained from smokers with or without COPD, and then stimulated with cigarette sm...
متن کاملComparison between cigarette smoke-induced emphysema and cigarette smoke extract-induced emphysema
BACKGROUND AND OBJECTIVE Emphysema is the main pathological feature of COPD and also is the focus of the related research. Although several emphysema animal models have been established, exact comparison of findings is seldom. The present study aimed to compare cigarette smoke (CS) exposure-induced emphysema model and intraperitoneal injection of cigarette smoke extract (CSE)-induced emphysema ...
